Yes, it had changed. When it began in 1952, it was a humor based suspense magazine similar to Tales From The Crypt. Hence it's original title "Tales Calculated To Drive You Mad". Then the sixties rolled along. The horror pushed to the distance and out of favor permanently, and Mad's comedy took on a much more counter culture bent. From wiki: "In the 1960s and beyond, it satirized such burgeoning topics as the sexual revolution, hippies, psychoanalysis, gun control, pollution, the Vietnam War and recreational drug use. The magazine gave equal time, generally negative, to counterculture drugs such as cannabis, and took a critical approach toward mainstream drugs such as tobacco and alcohol. Mad always satirized Democrats as mercilessly as it did Republicans. It also ran a good deal of less-topical material on such varied subjects as fairy tales, nursery rhymes, greeting cards, sports, small talk, poetry, marriage, comic strips,awards shows, cars and many other areas of general interest." It was culturally relevant and, in 1974, achieved a circulation of over two million copies per issue. However, the creator of Spy vs. Spy stopped drawing in the '80s and in the early '90s, Bill Gaines died and Don Martin went to Cracked. The material slid from observation to mere booger and fart jokes as teenagers and young adults no longer bought it. More wiki: "On April 1, 1997, the magazine publicized an alleged makeover, ostensibly designed to reach an older, more sophisticated readership. However, Salon 's David Futrelle opined that such content was very much a part of Mad's past." Since 2001, the mag has also featured paid advertising, where previously it only lampooned such things. So, in conclusion, Mad has actually undergone significant change over the past 50 years, despite Ebbomega's continued and stunning ignorance. I witnessed this change first hand as I still have many of their magazines ranging from reprints of their initial work through the '70s and many from the early '90s, but a simple trip to wikipedia would give you the same information.
