It depends. I personally prefer Harmonix games over Activision. I thought Guitar Hero 4 was one of the worst in the series.
If you have the original Rock band, go with Rock Band 2 for sure. They've gone and said that all the DLC is backwards compatible with Rock Band 2, as well as the songs from Rock Band 1 will be available on Rock Band 2. Pretty much this means about 200 songs when you start playing Rock Band.
Also, Rock Band 2 will have a new set of drums. There's some pictures floating around on the net, but they will be more drum like then the previous ones. Most important for you since I know you'd be interested in this:
Rock Band 2 will have drum lessons, with the end goal being that you actually learn to play drums. Drums pretty much are the only instrument that could work this way (Not so much musical theory as other instruments). So it could be really impressive.
Guitar Hero will have a make your own music part, which is cool I guess if you don't know how to play. Pretty much it's going to have the same features as Rock Band (make your own character etc...). Interesting thing to note is that they don't have a Guitar brand sponsor so you will be playing on guitars that you build yourself. You might like this or hate it. I love fender guitars so the choice is obvious for me.
Personally I would go for Rock Band 2 hands down. I hated Guitar Hero 4 and gave it away almost a day after I bought it.
